Our research group is interested in the cell biological basis of rare Mendelian diseases with highly tissue-specific manifestations, particularly those which disorder proteostasis (the process by which proteins are accurately synthesized, trafficked, and degraded). Our ongoing work focuses on diseases of protein aggregation in the brain and the lungs. We utilize a variety of experimental systems including cultured cells, organoids, and genetically engineered mice, applying tools including CRISPR/Cas9 genome editing, protein and small molecule mass spectrometry, RNA analysis, sequencing, confocal microscopy, basic biochemistry, and in vivo disease modeling.<p></p><p><b><br>
Job Summary</b></p>
The Chivukula Laboratory in the Center for Genomic Medicine (CGM) at Massachusetts General Hospital (MGH) is seeking a motivated, enthusiastic candidate to join our group as a research lab technician (RT).<br>

We involve RTs in all aspects of the scientific process: theory, experimental design, execution, interpretation, and figure preparation. Prior RTs have gone on to prestigious graduate, medical, and nursing school positions following their tenure and training in the laboratory.<br>

 In the Nakashima Lab at the Harvey Cushing Neuro-Oncology Laboratories, within the Department of Neurosurgery at Brigham and Women's Hospital, our research is focused on the developmental immunotherapy with tumor-permissive oncolytic viruses and tumor immune mechanisms governing highly immune tolerance and resistance in brain tumors. We use advanced DNA manipulation and animal models in conjunction with the latest molecular and immunological tools in our investigation of brain tumor and immunity. We are looking for an energetic and highly organized research assistant interested in various biological and immunological experiments, including plasmid DNA, RNA, genome extraction, (RT-)PCR, Western blot, immunohistochemistry, flow cytometry, virus, cell culture, and other essential molecular biological experiments.

We are looking for a Research Assistant to participate in various projects in the laboratory. Working under direct supervision of the principal investigator, the candidate will be responsible for: 1) handling live mice to set up crosses, wean litters, collect tissue biopsies for genotyping, and inject various drugs, 2) extracting, dissecting, processing and sectioning cochlear and brain tissues from euthanized mice, 3) detecting mRNA and proteins of interest by in situ hybridization and immunohistochemistry, 4) preparing buffers and reagents for molecular biology, 5) performing gel electrophoresis, 6) acquiring images using epifluorescence and confocal microscopy, 7) assisting in general laboratory maintenance, including setting up of equipment, and organizing reagents and supplies. In addition, opportunities will be available to conduct gene expression analysis using R or Python in collaborative projects.
