A full-time position is available for a highly motivated research technician at the translational immunogenomics laboratory (TIGL). The technician will contribute to new technologies for characterizing T cell specificity and neoantigen peptide properties in cancer and infectious disease. The laboratory uses various techniques in immunology, cell culture, molecular biology, protein engineering, and mass-spectrometry to develop and scale high throughput methods for mapping the antigen specificity of millions of T cells using single-cell RNA sequencing. TIGL also takes part in DFCI Neoantigen based cancer vaccine trials, and all the patient samples are analyzed in TIGL laboratories. The technician will be responsible for immune monitoring of ongoing Neoantigen-based cancer vaccine trials, protein purification, preparing samples for mass-spectrometry, and cellular immunology workflows, including flow cytometry and single-cell RNA seq. The technician should have excellent record-keeping skills and a drive to work develop new methods in immunology. Training is available in all protocols and procedures; experience with basic techniques in cell culture and biology is strongly preferred. Previous experience with computational biology, knowledge of R and single cell analysis is beneficial.</p><p><b><br>
Responsibilities:</b></p><p>
The candidate will be expected to independently carry out essential research functions in the laboratory that include but are not limited to experience in cell and molecular biology techniques.</p><p>

The Grabowski Laboratory is looking for a pre-medical student interested in translational research, or for a young investigator who seeks a medical research career to fill a position as a Research Technician II. The person in this job will carry out research on two major clinical problems: 1) the further development of a first effective therapy for a devastating thrombotic microangiopathy known as the childhood hemolytic uremic syndrome (eHUS), and 2) the study of von Willebrand Disease, the most common inherited bleeding disorder. Work for 2) encompasses laboratory microfluidics and biochemistry combined with clinical bleeding assessments and the obtaining of informed consent for research trials. We are presently examining the little understood role of the von Willebrand factor (VWF) pro-peptide in bleeding and clotting, and the function of VWF as an anti-angiogenic factor that, when deficient, may lead to unregulated angiogenesis and increased bleeding from fragile blood vessels in mucous membranes. Work for 1) involves the pursuit of novel therapeutic targets for eHUS by means of a genetically modified mouse model and an all-human endothelial cell culture model. The laboratory utilizes cell cultures of human endothelial cells, ELISAs for clotting factor activities, controlled hemodynamics of blood flow, fluorescence digital video-microscopy, real-time digital image processing (e.g., antigen co-localization) using a state-of-the-art imaging system, an ex vivo flow chamber to simulate flow in arteries, human arterioles, and MetaMorph Premier digital imaging software. This is a great opportunity for, as stated above, either a pre-medical student interested in translational research, or for a young investigator who seeks a medical research career. The lab director is a senior pediatric hematologist and an experienced scientist. Also overseeing the translational work is a senior medical hematologist. An applicant for this position should have a background in biology, chemistry, or a related other medical science.

The Research Assistant (RAI) works with the Laboratory Manager, Principal Investigators, and other staff to conduct screening and study visits. This position plays an essential role in the successful completion of multiple clinical research protocols being conducted in the Asthma Research Center at one time.<br>

The RAI is responsible for independently recruiting and screening study subjects for eligibility to participate in clinical trials. The RAI is also responsible for study data collection and entry, lab processing and maintaining study regulatory documents. They may be responsible for spearheading recruitment efforts for research studies. Excellent communication (oral and written), phone, and computer skills are necessary.<br>

Working under general supervision from the Laboratory Manager, provides support for clinical research studies.<br>

PRINCIPAL DUTIES AND RESPONSIBILITIES:<br>

1. Assists in the development and planning for the implementation of procedures for research studies evaluating asthma treatments and medications.<br>
<br>
2. Reviews, understands and obtains appropriate certifications for study specific protocols and ARC procedures.<br>
<br>
3. Actively contributes to the recruitment of study subjects for ARC studies. This includes assisting with developing and implementation of recruitment strategies, telephone screening of potential subjects, enlisting participation by scheduling appointments and sending out necessary paperwork.<br>
<br>
4. Performs study visits, including review of consent with subjects, following ARC procedures for study visits, and appropriate follow up with subjects.<br>
<br>
5. Clinical and pulmonary testing including performing, e.g., vital signs, EKG, blood draws, skin testing, spirometry, methacholine challenges, sputum induction, exercise challenges, assisting with certain procedures for bronchoscopies and other related procedures.<br>
<br>
6. Collects and reviews study data, ensuring compliance with protocol and data integrity. Refers appropriate clinical issues/questions to respective medical/managerial staff.<br>
<br>
7. Adheres to the administrative aspects of trials, including data quality, protocol violation reporting and adverse event reporting, and accurately and promptly maintains study related logs.<br>
<br>
8. Coordinates study data with Data Coordinating Center, corporate study sponsor or their CRO. Promptly resolves and responds to data queries.<br>

9. Prepares and presents data reports as directed.<br>
